HPLC OptimizationTM

This program optimizes isocratic and gradient HPLC conditions, as well as scale-up to preparative HPLC. It is based on relative resolution mapping from two initial gradient runs. Make two gradient runs at two different gradient times using the same starting and ending gradient. Input the dwell volume, flow rate, column length, mobile phase, starting and ending gradient percentage, number of compounds, the two gradient run times, peak names, retention times, and areas for both gradient run times.

This software is designed as an efficient laboratory tool for the following:

· Provide optimized isocratic mobile phase composition for your HPLC separation.
· Provide equivalent elution strength isocratic composition for other mobile phases.
· Provide optimized gradient conditions.
· Provide scale-up conditions for preparative HPLC.
· Provide optimized retention times.
· Provide a simulated chromatogram.
· Provide an estimate of the molecular weight of each compound.
· Provide a printed report of the results.
· Provide copy or paste data to and from a Windows spreadsheet program.

ISOCRATIC
This program provides the optimized isocratic solvent compositions for methanol, acetonitrile, and tetrahydrofuran, along with the optimized retention times, and the estimated molecular weight for each compound. The program will simulate what the chromatogram would look like. To see how the retention times change as you change the composition, just enter a new composition.

GRADIENT
This program provides the optimized gradient time, along with the optimized retention times, and the estimated molecular weight for each compound. The program will simulate what the chromatogram would look like. To see how the retention times change as you change the gradient time, just enter a new gradient time.

FINE TUNE
This program also provides the optimized gradient conditions after making only one gradient run. Enter the starting and ending gradient concentration in percentage (e.g., 5-100 %), the gradient run time (e.g., 30 min.), and the first and last retention time. The results for the new starting and ending gradient concentration and the new gradient run time will be displayed.

SCALE-UP
This program will scale-up from analytical to preparative HPLC. Enter the diameter of the analytical column, length, run time, and maximum loading. Then enter the diameter of the preparative column and length of the preparative column. The results for the calculated run time, maximum loading, calculated flow rate, and gradient delay time (hold at initial conditions) for the preparative column will be displayed.
