Quantikine^® mRNA is a microplate-based assay for the quantitation of cytokine-specific mRNA from total RNA samples using colorimetric detection. This assay allows quantitation of different targets on the same microplate and uses less sample RNA than a Northern blot or ribonuclease protection assay. In this assay, total RNA samples are hybridized with biotin-labeled capture oligonucleotides and digoxigenin-labeled detection oligonucleotides. The RNA-oligonucleotide hybrids are captured on a streptavidin-coated plate and are detected with anti-digoxigenin alkaline phophatase conjugate. The signal is then amplified using a substrate that generates a colorimetric end product.
