The columns operate on a negative selection principle, with monocytes, B cells, NK cells and granulocytes preferentially targeted for removal. Enriched T cells pass through the column and are recovered in the sterile effluent ready for tissue culture. Enrichment of cells via a negative cell selection strategy minimizes any potential activation of cells associated with positive selection techniques.
T cell columns have a wide range of loading capacities and are available in several sizes for processing variable numbers of cells. Columns enrich for either all CD3+ cells, or for subsets of T cells, such as: CD4+, CD8+, CD4+/CD45RA-, or CD4+/CD45RO- cells. T cell subset columns are supplied with an antibody cocktail designed to negatively deplete the cell population of no interest, thereby enriching for the desired T cell subset.
